Integrated omics approach reveals the molecular pathways activated in tomato by Kocuria rhizophila, a soil plant growth-promoting bacterium

Plant microbial biostimulants application has become a promising and eco-friendly agricultural strategy to improve crop yields, reducing chemical inputs for more sustainable cropping systems. The soil dwelling bacterium Kocuria rhizophila was previously characterized as Plant Growth Promoting Bacteria (PGPB) for its multiple PGP traits, such as indole-3-acetic acid production, phosphate solubilization capability and salt and drought stress tolerance. Here, we evaluated by a multi-omics approach, the PGP activity of K. rhizophila on tomato, revealing the molecular pathways by which it promotes plant growth. Transcriptomic analysis showed several up-regulated genes mainly related to amino acid metabolism, cell wall organization, lipid and secondary metabolism, together with a modulation in the DNA methylation profile, after PGPB inoculation. In agreement, proteins involved in photosynthesis, cell division, and plant growth were highly accumulated by K. rhizophila. Furthermore, “amino acid and peptides”, “monosaccharides”, and “TCA” classes of metabolites resulted the most affected by PGPB treatment, as well as dopamine, a catecholamine neurotransmitter mediating plant growth through S-adenosylmethionine decarboxylase (SAMDC), a gene enhancing the vegetative growth, up-regulated in tomato by K. rhizophila treatment. Interestingly, eight gene modules well correlated with differentially accumulated proteins (DAPs) and metabolites (DAMs), among which two modules showed the highest correlation with nine proteins, including a nucleoside diphosphate kinase, and cytosolic ascorbate peroxidase, as well as with several amino acids and metabolites involved in TCA cycle. Overall, our findings highlighted that sugars and amino acids, energy regulators, involved in tomato plant growth, were strongly modulated by the K. rhizophila-plant interaction.

Simultaneously enhancing plant growth and immunity through the application of engineered Bacillus subtilis expressing a microbial pattern1

Simultaneously enhancing plant growth and disease resistance is an ideal goal in Agriculture. Significant efforts have been made to promote plant growth or immunity through the use of biological reagents, such as the application of beneficial microbes and plant immunity inducers. However, balancing plant immunity and growth remains a challenging task. In this study, we engineered the plant growth-promoting bacterium Bacillus subtilis OKB105 to express a secreted microbial pattern, flg22, and accessed its activity in enhancing both plant growth and disease resistance. The OKB105 (flg22) strain exhibited plant growth-promoting activity similar to the OKB105 strain containing an empty vector, OKB105 (EV). Furthermore, the OKB105 (flg22) strain significantly enhanced plant resistance against two distinct pathogens, Pseudomonas syringae DC3000 ΔhopQ1-1 and Phytophthora parasitica, compared to OKB105 (EV), confirming that the engineered OKB105 (flg22) effectively enhances plant disease resistance. Interestingly, root irrigation with OKB105 (flg22) also markedly boosted the plant’s aboveground resistance to pathogens compared to OKB105 (EV). We further demonstrated that OKB105 (flg22) can be applied to confer resistance to pathogens in other plants that recognize flg22. Finally, RNA-Seq and qRT-PCR analyses illustrated that OKB105 (flg22) effectively induced the expression of defense-related genes in pattern-triggered immunity. Our results prove that employing an engineered beneficial microbe expressing a microbial pattern is a promising strategy for simultaneously enhancing plant growth and immunity.

Understanding the dynamic interactions of root-knot nematodes and their host: role of plant growth promoting bacteria and abiotic factors

Root-knot nematodes (Meloidogyne spp., RKN) are among the most destructive endoparasitic nematodes worldwide, often leading to a reduction of crop growth and yield. Insights into the dynamics of host-RKN interactions, especially in varied biotic and abiotic environments, could be pivotal in devising novel RKN mitigation measures. Plant growth-promoting bacteria (PGPB) involves different plant growth-enhancing activities such as biofertilization, pathogen suppression, and induction of systemic resistance. We summarized the up-to-date knowledge on the role of PGPB and abiotic factors such as soil pH, texture, structure, moisture, etc. in modulating RKN-host interactions. RKN are directly or indirectly affected by different PGPB, abiotic factors interplay in the interactions, and host responses to RKN infection. We highlighted the tripartite (host-RKN-PGPB) phenomenon with respect to (i) PGPB direct and indirect effect on RKN-host interactions; (ii) host influence in the selection and enrichment of PGPB in the rhizosphere; (iii) how soil microbes enhance RKN parasitism; (iv) influence of host in RKN-PGPB interactions, and (v) the role of abiotic factors in modulating the tripartite interactions. Furthermore, we discussed how different agricultural practices alter the interactions. Finally, we emphasized the importance of incorporating the knowledge of tripartite interactions in the integrated RKN management strategies.

Improvement of morphophysiological and anatomical attributes of plants under abiotic stress conditions using plant growth-promoting bacteria and safety treatments.

Drought and salinity are the major abiotic stress factors negatively affecting the morphophysiological, biochemical, and anatomical characteristics of numerous plant species worldwide. The detrimental effects of these environmental factors can be seen in leaf and stem anatomical structures including the decrease in thickness of cell walls, palisade and spongy tissue, phloem and xylem tissue. Also, the disintegration of grana staking, and an increase in the size of mitochondria were observed under salinity and drought conditions. Drought and salt stresses can significantly decrease plant height, number of leaves and branches, leaf area, fresh and dry weight, or plant relative water content (RWC%) and concentration of photosynthetic pigments. On the other hand, stress-induced lipid peroxidation and malondialdehyde (MDA) production, electrolyte leakage (EL%), and production of reactive oxygen species (ROS) can increase under salinity and drought conditions. Antioxidant defense systems such as catalase, peroxidase, glutathione reductase, ascorbic acid, and gamma-aminobutyric acid are essential components under drought and salt stresses to protect the plant organelles from oxidative damage caused by ROS. The application of safe and eco-friendly treatments is a very important strategy to overcome the adverse effects of drought and salinity on the growth characteristics and yield of plants. It is shown that treatments with plant growth-promoting bacteria (PGPB) can improve morphoanatomical characteristics under salinity and drought stress. It is also shown that yeast extract, mannitol, proline, melatonin, silicon, chitosan, α-Tocopherols (vitamin E), and biochar alleviate the negative effects of drought and salinity stresses through the ROS scavenging resulting in the improvement of plant attributes and yield of the stressed plants. This review discusses the role of safety and eco-friendly treatments in alleviating the harmful effects of salinity and drought associated with the improvement of the anatomical, morphophysiological, and biochemical features in plants.

Using multi-omics to explore the effect of Bacillus velezensis SAAS-63 on resisting nutrient stress in lettuce

To avoid the unreasonable use of chemical fertilizer, an environmentally friendly means of improving soil fertility is required. This study explored the role of the plant growth-promoting rhizosphere bacteria (PGPR) strain Bacillus velezensis SAAS-63 in improving nutrient stress in lettuce. Compared with no inoculation, B. velezensis SAAS-63 inoculants exhibited significantly increased fresh weight, root length, and shoot height under nutrient deficiency, as well as improved antioxidant activities and proline contents. The exogenous addition of B. velezensis SAAS-63 also significantly increased the accumulation of macroelements and micronutrients in lettuce. To elucidate the resistance mechanisms induced by B. velezensis SAAS-63 under nutrient stress, high-throughput sequencing and multi-omics analysis were performed. Inoculation with B. velezensis SAAS-63 altered the microbial community of the rhizosphere and increased the relative abundances of Streptomyces, Actinoallomurus, Verrucomicrobia, and Chloroflexi. It is worth noting that the inoculant SAAS-63 can affect plant rhizosphere metabolism. The inoculant changed the metabolic flow of phenylpropanoid metabolic pathway under nutrient deficiency and promoted phenylalanine to participate more in the synthesis of lignin precursors and coumarin substances by inhibiting the synthesis of flavone and isoflavone, thus improving plant resistance. This study showed that the addition of inoculant SAAS-63 could help plants recruit microorganisms to decompose and utilize trehalose and re-established the carbon metabolism of the plant rhizosphere. Additionally, microbes were found to be closely related to the accumulation of metabolites based on correlation analysis. The results indicated that the addition of PGPRs has an important role in regulating soil rhizosphere microbes and metabolism, providing valuable information for understanding how PGPRs affect complex biological processes and enhance plant adaptation to nutrient deficiency.Key points• Inoculation with SAAS-63 significantly promoted plant growth under nutrient-deficient conditions• Inoculation with SAAS-63 affected rhizosphere microbial diversity and community structure• Inoculation with SAAS-63 affected plant rhizosphere metabolism and induced plants to synthesize substances that resist stressGraphical abstract

Plant Growth-Promoting Bacteria Influence Microbial Community Composition and Metabolic Function to Enhance the Efficiency of Hybrid pennisetum Remediation in Cadmium-Contaminated Soil

The green and efficient remediation of soil cadmium (Cd) is an urgent task, and plant-microbial joint remediation has become a research hotspot due to its advantages. High-throughput sequencing and metabolomics have technical advantages in analyzing the microbiological mechanism of plant growth-promoting bacteria in improving phytoremediation of soil heavy metal pollution. In this experiment, a pot trial was conducted to investigate the effects of inoculating the plant growth-promoting bacterium Enterobacter sp. VY on the growth and Cd remediation efficiency of the energy plant Hybrid pennisetum. The test strain VY-1 was analyzed using high-throughput sequencing and metabolomics to assess its effects on microbial community composition and metabolic function. The results demonstrated that Enterobacter sp. VY-1 effectively mitigated Cd stress on Hybrid pennisetum, resulting in increased plant biomass, Cd accumulation, and translocation factor, thereby enhancing phytoremediation efficiency. Analysis of soil physical-chemical properties revealed that strain VY-1 could increase soil total nitrogen, total phosphorus, available phosphorus, and available potassium content. Principal coordinate analysis (PCoA) indicated that strain VY-1 significantly influenced bacterial community composition, with Proteobacteria, Firmicutes, Chloroflexi, among others, being the main differential taxa. Redundancy analysis (RDA) revealed that available phosphorus, available potassium, and pH were the primary factors affecting bacterial communities. Partial Least Squares Discriminant Analysis (PLS-DA) demonstrated that strain VY-1 modulated the metabolite profile of Hybrid pennisetum rhizosphere soil, with 27 differential metabolites showing significant differences, including 19 up-regulated and eight down-regulated expressions. These differentially expressed metabolites were primarily involved in metabolism and environmental information processing, encompassing pathways such as glutamine and glutamate metabolism, α-linolenic acid metabolism, pyrimidine metabolism, and purine metabolism. This study utilized 16S rRNA high-throughput sequencing and metabolomics technology to investigate the impact of the plant growth-promoting bacterium Enterobacter sp. VY-1 on the growth and Cd enrichment of Hybrid pennisetum, providing insights into the regulatory role of plant growth-promoting bacteria in microbial community structure and metabolic function, thereby improving the microbiological mechanisms of phytoremediation.

Endophytic Bacillus pumilus G5 Interacting with Silicon to Improve Drought Stress Resilience in Glycyrrhiza uralensis Fisch. by Modulating Nitrogen Absorption, Assimilation, and Metabolism Pathways.

Drought stress has become the primary severe threat to global agriculture production, including medicinal plants. Plant growth-promoting bacteria (PGPB) and environmentally friendly element silicon (Si) have emerged as effective methods in alleviating drought stress in various plants. Here, the effects of the plant endophytic G5 interaction with Si on regulating nitrogen absorption, assimilation, and metabolism pathways were investigated in the morphophysiological and gene attributes of Glycyrrhiza uralensis exposed to drought. Results showed that G5+Si application improved nitrogen absorption and assimilation by increasing the available nitrogen content in the soil, further improving the nitrogen utilization efficiency. Then, G5+Si triggered the accumulation of the major adjustment substances proline, γ-aminobutyric acid, putrescine, and chlorophyll, which played an important role in contributing to maintaining balance and energy supply in G. uralensis exposed to drought. These findings will provide new ideas for the combined application of PGPR and Si on both soil and plant systems in a drought habitat.

Diversity of Multi-Trait Plant Growth-Promoting Culturable Bacteria from Natural Geothermal Springs

Natural geothermal springs are extreme ecological niches harboring a variety of microbial resources with potential roles in agriculture. The objective of the current study was to investigate the functional microbial diversity of Tattapani and Sohna hot springs. A total of 19 best bacterial strains were selected based on multifunctional plant growth-promoting (PGP) properties. The majority of selected bacterial strains possessed antagonistic activity and were able to solubilize zinc (2.58- 6.00 ZSI), potassium (2.11- 4.00 KSI), phosphate (3.00 PSI) and silicate (1.50- 4.00 SiSI) under in-vitro conditions. Out of selected strains 84% and 73% strains showed siderophore (20.08 to 70.15% siderophore units) and Indole-3-acetic acid production (20.50 − 37.15 μg ml−1), respectively. 16S rDNA sequencing of selected strains showed the predominance of Bacillaceae (44%), Pseudomonadaceae (22%), Moraxellaceae (11%), Enterobacteriaceae (11%), Paracoccaceae (6%) and Acidithiobacillaceae (6%). The outcome of the current investigation shows that multitrait plant growth-promoting bacteria could be isolated from the hot springs and may have a potential impact on crop production of problematic soils. Therefore, these potential strains can be further harnessed for preparation of bioformulations for the sustainable agriculture.

Response strategies of slash pine (Pinus elliottii) to cadmium stress and the gain effects of inoculation with Herbaspirillum sp. YTG72 in alleviating phytotoxicity and enhancing accumulation of cadmium.

Phytoremediation using fast-growing woody plants assisted by plant growth-promoting bacteria (PGPB) on cadmium (Cd)-contaminated sites is considered a promising technique; however, its remediation efficiency is still affected by multiple factors. In this study, the mining areas' soil conditions were simulated with different Cd addition levels (0, 3, 6, 9mgkg-1) in order to investigate the response strategy to Cd stress of fast-growing economic tree species, slash pine (Pinus elliottii), and the effects of inoculation with the PGPB strain Herbaspirillum sp. YTG72 on the physiological activity and Cd accumulation of plants. The main results showed that there were significant (p < 0.05) increases in contents of chlorophyll and nutrient elements (P, K, Ca, and Mg) at low Cd addition level (3mgkg-1) compared to non-Cd addition treatment. When the additive amount of Cd increased, the growth of plants was severely inhibited and the content of proline was increased, as well as Cd in plants. Besides, the ratios of K:P, Ca:P, and Mg:P in plants were negatively correlated with the contents of Cd in plants and soils. Inoculation of P. elliottii with the PGPB strain Herbaspirillum sp. YTG72 improved the physiological functions of the plants under Cd stress and activated the antioxidant system, reduced the accumulation of proline, and decreased the ratios of K:P, Ca:P, and Mg:P in plant. More importantly, planting P. elliottii in Cd-contaminated soil could significantly (p < 0.05) reduce the Cd content in the rhizosphere soil, and furthermore, inoculation treatment could promote the reduction of soil Cd content and increased the accumulation of Cd by root. The results of the present study emphasized the Cd response mechanism of P. elliottii based on multifaceted regulation, as well as the feasibility of strain Herbaspirillum sp. YTG72 assisted P. elliottii for the remediation on Cd-contaminated sites.

Asp-tRNAAsn/Glu-tRNAGln amidotransferase A subunit-like amidase mediates the degradation of insecticide flonicamid by Variovorax boronicumulans CGMCC 4969

The main metabolic product of the pyridinecarboxamide insecticide flonicamid, N-(4-trifluoromethylnicotinyl)glycinamide (TFNG-AM), has been shown to have very high mobility in soil, leading to its accumulation in the environment. Catabolic pathways of flonicamid have been widely reported, but few studies have focused on the metabolism of TFNG-AM. Here, the rapid transformation of TFNG-AM and production of the corresponding acid product N-(4-trifluoromethylnicotinoyl) glycine (TFNG) by the plant growth-promoting bacterium Variovorax boronicumulans CGMCC 4969 were investigated. With TFNG-AM at an initial concentration of 0.86 mmol/L, 90.70 % was transformed by V. boronicumulans CGMCC 4969 resting cells within 20 d, with a degradation half-life of 4.82 d. A novel amidase that potentially mediated this transformation process, called AmiD, was identified by bioinformatic analyses. The gene encoding amiD was cloned and expressed recombinantly in Escherichia coli, and the enzyme AmiD was characterized. Key amino acid residue Val154, which is associated with the catalytic activity and substrate specificity of signature family amidases, was identified for the first time by homology modeling, structural alignment, and site-directed mutagenesis analyses. When compared to wild-type recombinant AmiD, the mutant AmiD V154G demonstrated a 3.08-fold increase in activity toward TFNG-AM. The activity of AmiD V154G was greatly increased toward aromatic L-phenylalanine amides, heterocyclic TFNG-AM and IAM, and aliphatic asparagine, whereas it was dramatically lowered toward benzamide, phenylacetamide, nicotinamide, acetamide, acrylamide, and hexanamid. Quantitative PCR analysis revealed that AmiD may be a substrate-inducible enzyme in V. boronicumulans CGMCC 4969. The mechanism of transcriptional regulation of AmiD by a member of the AraC family of regulators encoded upstream of the amiD gene was preliminarily investigated. This study deepens our understanding of the mechanisms of metabolism of toxic amides in the environment, providing new ideas for microbial bioremediation.

Pilot-scale production of Bacillus subtilis MSCL 897 spore biomass and antifungal secondary metabolites in a low-cost medium.

Bacillus subtilis is a plant growth promoting bacterium (PGPB) that acts as a microbial fertilizer and biocontrol agent, providing benefits such as boosting crop productivity and improving nutrient content. It is able to produce secondary metabolites and endospores simultaneously, enhancing its ability to survive in unfavorable conditions and eliminate competing microorganisms. Optimizing cultivation methods to produce B. subtilis MSCL 897 spores on an industrial scale, requires a suitable medium, typically made from food industry by-products, and optimal temperature and pH levels to achieve high vegetative cell and spore densities with maximum productivity. This research demonstrates successful pilot-scale (100L bioreactor) production of a biocontrol agent B. subtilis with good spore yields (1.5×109 spores mL-1) and a high degree of sporulation (>80%) using a low-cost cultivation medium. Culture samples showed excellent antifungal activity (1.6-2.3cm) against several phytopathogenic fungi. An improved methodology for inoculum preparation was investigated to ensure an optimal seed culture state prior to inoculation, promoting process batch-to-batch repeatability. Increasing the molasses concentration in the medium and operating the process in fed-batch mode with additional molasses feed, did not improve the overall spore yield, hence, process operation in batch mode with 10g molasses L-1 is preferred. Results also showed that the product quality was not significantly impacted for up to 12months of storage at room temperature. An economically-feasible process for B. subtilis-based biocontrol agent production was successfully developed at the pilot scale.

Assessment of Plant-Growth Promoting Potential of Bacteria Isolated from Amazonian Black Pepper Roots

AbstractPurpose: In the Amazon, farmers use chemical fertilizers on a large scale to meet the nutritional requirements of some crops. Plant-growth promoting bacteria (PGPB) offer a sustainable alternative to enhance crop productivity. This study aimed to prospect novel PGPB from Amazonian black pepper (Piper nigrum L.) roots. Methods: Bacterial isolates were obtained from plant roots, evaluated for their biofertilizing potential, and the most promising strain was selected for genome sequencing. Taxonomic classification was based on 16 S rRNA gene sequencing. ACC deaminase activity, phosphate solubilization, and nitrogen fixation were assessed. Genome sequencing was performed using the Ion GeneStudio S5 platform. Results: The 20 isolates were affiliated to Enterobacter (7 isolates), Klebsiella (4 isolates), Kosakonia (5 isolates), Bacillus (2 isolates), and two unclassified bacteria. Seven isolates were positive for ACC deaminase activity, while four were positive for the presence of the nifH gene. Nitrogenase gene was found only in Kosakonia isolates. Ninety isolates were able to solubilize phosphate. The isolate Pn16 was the most promising and presented a genome of 6,432,985 bp, GC content of 55%, 6,465 Coding Sequences, 10 Symbiotic Islands, 28 biosynthetic gene clusters, and several genes involved in plant-growth promotion such as phoU-pstSCAB-phoBR, oqxAB, ipdC, speADEGF, nifHDK. Conclusions: We were able to isolate a bacterium with potential for biofertilization. Based on phylogeny and Average Nucleotide Identity, we propose the classification of the Pn16 isolate as Kosakonia pseudosacchari Pn16.

The Effects of Plant Growth-promoting Bacteria on Seed Germination and Seedling Growth in Crassocephalum rubens

Crassocephalum rubens, an herb with potential anticancer properties, encounters challenges in production efficiency due to small seed size and elevated temperatures in cultivation areas. Our study investigates the effects of seed pelleting and plant growth-promoting bacteria (PGPB) on germination and seedling growth across temperature variations in Taiwan. Pelleted seeds demonstrate superior germination percentages, linked to enhanced seed quality. Elevated temperature, particularly at 30/25°C, enhanced germination performance, with the highest final germination percentage observed in pelleted seeds treated with water. Unexpectedly, PGPB strains – Klebsiella sp. KsGRB10 and Bacillus sp. BsGFB04 – exhibited limited impact on germination rates, showing a marginal increase in seedling growth under 25/20°C and 30/25°C, respectively. Air temperature fluctuations influenced seedling vigor, leaf color, and physiological parameters. Remarkably, inoculation with BsGFB04 and KsGRB10 enhanced C. rubens’ tolerance to high-temperature stress conditions. Diurnal measurements in week 4, under 25/20°C, reveal that PGPB inoculation decreased stomatal conductance and transpiration rate while maintaining the quantum yield of PSII, indicating potentially improved water-use efficiency. This study provides crucial insights into the interplay among PGPB, environmental stress, and the physiology of a wild species, paving the way for further research in the domestication of C. rubens for medicinal herb mass production.

Enhancing Pisum sativum growth and symbiosis under heat stress: the synergistic impact of co-inoculated bacterial consortia and ACC deaminase-lacking Rhizobium

The 1-aminocyclopropane-1-carboxylate (ACC) deaminase is a crucial bacterial trait, yet it is not widely distributed among rhizobia. Hence, employing a co-inoculation approach that combines selected plant growth-promoting bacteria with compatible rhizobial strains, especially those lacking ACC deaminase, presents a practical solution to alleviate the negative effects of diverse abiotic stresses on legume nodulation. Our objective was to explore the efficacy of three non-rhizobial endophytes, Phyllobacterium salinisoli (PH), Starkeya sp. (ST) and Pseudomonas turukhanskensis (PS), isolated from native legumes grown in Tunisian arid regions, in improving the growth of cool-season legume and fostering symbiosis with an ACC deaminase-lacking rhizobial strain under heat stress. Various combinations of these endophytes (ST + PS, ST + PH, PS + PH, and ST + PS + PH) were co-inoculated with Rhizobium leguminosarum 128C53 or its ΔacdS mutant derivative on Pisum sativum plants exposed to a two-week heat stress period.Our findings revealed that the absence of ACC deaminase activity negatively impacted both pea growth and symbiosis under heat stress. Nevertheless, these detrimental effects were successfully mitigated in plants co-inoculated with ΔacdS mutant strain and specific non-rhizobial endophytes consortia. Our results indicated that heat stress significantly altered the phenolic content of pea root exudates. Despite this, there was no impact on IAA production. Interestingly, these changes positively influenced biofilm formation in consortia containing the mutant strain, indicating synergistic bacteria-bacteria interactions. Additionally, no positive effects were observed when these endophytic consortia were combined with the wild-type strain. This study highlights the potential of non-rhizobial endophytes to improve symbiotic performance of rhizobial strains lacking genetic mechanisms to mitigate stress effects on their legume host, holding promising potential to enhance the growth and yield of targeted legumes by boosting symbiosis.

Two microbes assisting Miscanthus floridulus in remediating multi-metal(loid)s-contaminated soil.

Miscanthus has good tolerance to multi-metal(loid)s and has received increasing attention in remediated studies of metal(loid)s-contaminated soil. In this study, we conducted phytoextraction techniques to investigate the synergic effects of remediation of multi-metal(loid)s-contaminated soil by Miscanthus floridulus (Lab.) and two plant growth-promoting bacteria (PGPB), TS8 and MR2, affiliated to Enterobacteriaceae. The results exhibited a decrease of arsenic (15.27-21.50%), cadmium (8.64-15.52%), plumbum (5.92-12.76%), and zinc (12.84-24.20%) except for copper contents in the soil in bacterial inoculation groups, indicating that MR2 and TS8 could enhance the remediation of metal(loid)s. Moreover, increased fresh/dry weight and height indicated that inoculated bacteria could promote Miscanthus growth. Although the activities of antioxidant enzymes and the content of chlorophyll in the overground tissues showed no significant increase or even decrease, the activities of antioxidant enzymes in the underground tissues and soil were elevated by 48.95-354.17%, available P by 19.07-23.02%, and available K by 15.34-17.79% (p < 0.05). Bacterial inoculants could also decrease the soil pH. High-throughput sequencing analysis showed that the bacterial inoculant affected the rhizosphere bacterial community and reduced community diversity, but the relative abundance of some PGPB was found to increase. Phylogenetic molecular ecological networks indicated that bacterial inoculants reduced interactions between rhizosphere bacteria and thereby led to a simpler network structure but increased the proportion of positive-correlation links and enhanced the metabiosis and symbiosis of those bacteria. Spearman's test showed that OTUs affiliated with Enterobacteriaceae and soil nutrients were critical for metal(loid) remediation and Miscanthus growth. The results of this study provide a basis for the synergic remediation of multi-metal(loid)s-contaminated soils by Miscanthus and PGPB and provide a reference for the subsequent regulation of Miscanthus remediation efficiency by the other PGPB or critical bacteria.

Amelioration of growth of maize (Zea mays L.) seedling using plant growth promoting bacteria

This research was aimed to screen plant growth promoting bacteria (PGPB) from soil and study its effect on maize plant growth. PGPB were isolated from Saurashtra coastal region soil and cultured in Nitrogen fixing Ashby’s medium to find potent of PGPBs, we conducted a thorough screening process, assessed their abilities in phosphate and zinc solubilization, siderophore production, hydrogen cyanide (HCN) release and the antifungal activity was performed against Fusarium oxysporum, a pathogenic fungus. These tests helped us identify bacteria with plant growth-promoting characteristics for plants. Bacterial isolates which provided better results were sequenced and sequences were submitted to NCBI. Bacterial isolates selected for application on maize in primary screening showed most treated seeds increased the seedling vigor of maize. In the latter stages of screening where bacterial consortia were developed from primary and secondary screening. In 30 days, the experiment in maize plant height, number of leaves, chlorophyll content and anatomy was analysed. All the bacterium consortiums displayed an increase in height (24.75%), number of leaves (47.77%) and total chlorophyll content (23.59%) as compared to the control maize plant. Additionally, microscopic examination of the treated plants showed improved growth, especially in the increased starch grain content in the leaves, stems and roots. Out of the eleven PGPB consortia, 3 specific PGPB consortia in this study have significantly substantiated the growth of maize plants as evidenced by the comprehensive analysis of anatomical features.

Unveiling stress-adapted endophytic bacteria: Characterizing plant growth-promoting traits and assessing cross-inoculation effects on Populus deltoides under abiotic stress

In the face of the formidable environmental challenges precipitated by the ongoing climate change, Plant Growth-Promoting Bacteria (PGPB) are gaining widespread acknowledgement for their potential as biofertilizers, biocontrol agents, and microbial inoculants. However, a knowledge gap pertains to the ability of PGPB to improve stress tolerance in forestry species via cross-inoculation. To address this gap, the current investigation centres on PGPBs, namely, Acinetobacter johnsonii, Cronobacter muytjensii, and Priestia endophytica, selected from the phyllosphere of robust and healthy plants thriving in the face of stress-inducing conditions. These strains were selected based on their demonstrated adaptability to saline, arid, and nitrogen-deficient environments. The utilization of PGPB treatment resulted in an improvement of stomatal conductance (gs) and transpiration rate (E) in poplar plants exposed to both salt and drought stress. It also induced an increase in essential biochemical components such as proline (PRO), catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD). These reactions were accompanied by a decrease in leaf malonaldehyde (MDA) content and electrolyte leakage (EL). Furthermore, the PGPB treatment demonstrated a notable enhancement in nutrient absorption, particularly nitrogen and carbon, achieved through the solubilization of nutrients. The estimation of canopy temperature via thermal imaging proved to be an efficient method for distinguishing stress reactions in poplar than conventional temperature recording techniques. In summation, the utilization of PGPB especially Cronobacter muytjensii in this study, yielded profound improvements in the stress tolerance of poplar plants, manifesting in reduced membrane lipid peroxidation, enhanced photosynthesis, and bolstered antioxidant capacity within the leaves.

Screening Plant Growth-Promoting Bacteria with Antimicrobial Properties for Upland Rice.

This study explores beneficial bacteria isolated from the roots and rhizosphere soil of Khao Rai Leum Pua Phetchabun rice plants. A total of 315 bacterial isolates (KK1 to KK315) were obtained. Plant growth-promoting traits (phosphate solubilization and indole-3-acetic acid (IAA) production), and antimicrobial activity against three rice pathogens (Curvularia lunata NUF001, Bipolaris oryzae 2464, and Xanthomonas oryzae pv. oryzae) were assessed. KK074 was the most prolific in IAA production, generating 362.6±28.0 µg/ml, and KK007 excelled in tricalcium phosphate solubilization, achieving 714.2±12.1 µg/ml. In antimicrobial assays using the dual culture method, KK024 and KK281 exhibited strong inhibitory activity against C. lunata, and KK269 was particularly effective against B. oryzae. In the evaluation of antimicrobial metabolite production, KK281 and KK288 exhibited strong antifungal activities in cell-free supernatants. Given the superior performance of KK281, taxonomically identified as Bacillus sp. KK281, it was investigated further. Lipopeptide extracts from KK281 had significant antimicrobial activity against C. lunata and a minimum inhibitory concentration (MIC) of 3.1 mg/ml against X. oryzae pv. oryzae. LC-ESI-MS/MS analysis revealed the presence of surfactin A in the lipopeptide extract. The crude extract was non-cytotoxic to the L-929 cell line at tested concentrations. In conclusion, the in vitro plant growth-promoting and disease-controlling attributes of Bacillus sp. KK281 make it a strong candidate for field evaluation to boost plant growth and manage disease in upland rice.

Effect of Plant Growth-Promoting Bacteria (PGPB) on the Development of Pea crop (Pisum sativum L.)

Microorganisms are of great importance in agriculture in terms of plant nutrients by reducing the need for chemical fertilization. In recent years, plant growth-promoting bacteria (PGPB) have been widely used as biological fertilizers (BF) in agriculture. This study was conducted to determine the effect of plant growth-promoting bacteria on the development of pea plants. Firstly the phosphate solubilization and nitrogen fixation potentials of the bacteria used in this study were determined. In the study, the effects of 4 different combinations, F1 [(Rhizobium sp. (FR-13) and Pseudomonas alcaligenes (FDG121)], F2 [(Pseudomonas fluorescens biotype F (FDG-7), Rhizobium sp. (FR-18) and Bacillus-megaterium-GC subgroup B(FDG-134)], F3 [Arthrobacter oxydans (FDG-72), Bacillus-megaterium-GC subgroup B (FDG-146), Rhizobium sp. (FR-11)] and F4 [Acinetobacter genospecies 9 (FDG-116), Brevibacillus agri (FDG-118), Methylobacterium zatmanii (FDG-123) and Bacillus-megaterium-GC subgroup A (FDG-153)] were investigated. Formulations made with bacteria that were found to be the best in terms of the properties specified among these strains were tested against pea plants under greenhouse conditions and their effects on the plant's total fresh and dry weight were investigated. The study was set up to have 3 replications. As a result of the statistical analysis made with the data obtained, the formulations used compared to the control; F2, F3 and F1 applications were important in total fresh weight, respectively, and F2 and F3 applications were important in total dry weight. As a result, these 3 formulations are especially effective on the yield of pea plants and can be used as potential biofertilizers.
